چکیده

PURPOSE: The hypoxia-signaling pathway regulates endochondral ossification, and is critically important for proper modulation of inflammation, angiogenesis, and osteogenesis following trauma. Although hypoxia inducible factor-1α (Hif-1α) and hypoxia inducible factor-2α (Hif2α) are critical mediators of this pathway, the role of these factors in fracture healing remains unclear, particularly with regards to the regulation of inflammation after injury. In this study, we define the role of hypoxia signaling in fracture healing, with the ultimate goal of identifying novel pharmacologic targets to improve fracture healing in a targeted manner.

Notch Signaling is Enhanced During Endochondral Bone Regeneration Relative to Intramembranous Regeneration

INTRODUCTION: Bone regeneration occurs through a series of spatiotemporal events influenced by the mechanical environment, various local and systemic factors, and the embryological origin of the specific bone. Long bone fractures treated with flexible fixation heal primarily through endochondral ossification. Following an initial inflammatory phase, mesenchymal cells proliferate and undergo cho...

Regulation of Bone Metabolism

Bone is formed through the processes of endochondral and intramembranous ossification. In endochondral ossification primary mesenchymal cells differentiate to chondrocytes and then are progressively substituted by bone, while in intramembranous ossification mesenchymal stem cells (MSCs) differentiate directly into osteoblasts to form bone. The steps of osteogenic proliferation, differentiation,...

Prolyl Hydroxylase Domain-Containing Protein 2 (Phd2) Regulates Chondrocyte Differentiation and Secondary Ossification in Mice

Endochondral ossification plays an important role in the formation of the primary ossification centers (POCs) and secondary ossification centers (SOCs) of mammalian long bones. However, the molecular mechanisms that regulate POC and SOC formation are different. We recently demonstrated that Prolyl Hydroxylase Domain-containing Protein 2 (Phd2) is a key mediator of vitamin C effects on bone. We ...

Hypoxia Enhances Chondrogenesis and Prevents Terminal Differentiation through PI3K/Akt/FoxO Dependent Anti-Apoptotic Effect

Hypoxia, a common environmental condition, influences cell signals and functions. Here, we compared the effects of hypoxia (1% oxygen) and normoxia (air) on chondrogenic differentiation of human mesenchymal stem cells (MSCs). For in vitro chondrogenic differentiation, MSCs were concentrated to form pellets and subjected to conditions appropriate for chondrogenic differentiation under normoxia a...
